One way that we use is to unpack the firefox executables into a directory, this gives you setup.exe and 3 folders. The key folder to modify is localized. AFAIK this is static between major revision. With each new revision you need to unpack and overwrite the localized files with your own modifications.
Note that the correct way to do this is, according to Mozilla, is to create you own localization (en-COMPANY) and deploy it. However I cant justify making 5 or 6 prefs changes and a dictionary change as a whole new deployment so I just edit the default en-US
